You connected a sound card on the client or on the server ?

Did you check if your usb sound card is recognized by Debian systems. Maybe it is not a piserver problem. Maybe you need a driver. Also, I don't understand why you need to connect a usb sound card to the client. You can get the sound through the hdmi connector...

My application requires a microphone and speakers, which are standard for every operating system. Of course the Debian server recognizes a USB. Sound card

I am using Piserver since it was created and I never tried to connect anything else than mouse an keyboards. I don't think it is advisable to do what you try to do (but I may be wrong). Not only, you need to have your devices to be recognized by the os but you also have to deal with permissions problems. Keep in mind that you cannot have root access on the client, the changes have to be done on the server in the terminal of the os served to the client.
